Сегодня принципы 3R, «принципы гуманной методики эксперимента», являются общепринятым мировым стандартом, который позволяет в значительной степени сократить число лабораторных животных. При этом очень важно, чтобы сокращение числа животных не противоречило интересам исследования и не снижало достоверности полученных экспериментальных данных. Анализируются основные подходы к определению объема выборки экспериментальных животных при разном уровне доступности предварительных сведений об изучаемом лекарственном средстве. Даны примеры использования методов «Power and Sample Size Analysis» и «выравнивания ресурса» как способов оценки достаточности выборки экспериментальных животных. Рассмотрены основные подходы к выбору количества животных при изучении острой токсичности по некоторым зарубежным и российским руководствам. Проанализирован метод расчета объема выборки в случае достаточного количества предварительных экспериментальных данных на примере оценки биохимических параметров в ходе исследования хронической токсичности. Систематизированы рекомендации зарубежных агентств, регулирующих обращение лекарственных средств, по выбору числа экспериментальных животных в зависимости от длительности исследования, возраста животных к началу исследования и других факторов.
